It would likely be the same as doing the Z32 transmission depending on the starter. If the starter is mounted next to the block that might make it a bit more difficult.
Any particular reason you want to go with that transmission? Just for the 6 speed? Reason I ask is the TT transmission is proven to be very strong, it has wider gearing which if you go boosted will make for a much faster setup but with the VH's broad torque band will likely be quicker anyways, and finally the 6th gear of the S2K transmission is a higher ratio than the 5th of the TT transmission (just like the 350Z 6-speed). Your cruising rpm's will be higher in 6th gear than in 5th with the TT transmission so you'll likely lose a good deal of fuel economy.
I'd venture to guess that the S2K 6-speed likely won't take the abuse of the VH for too long, especially if modified a bit.
If you absolutely want to have a 6-speed, the T56 out of a Cobra would likely be the best. They are also in Camaro's/Firebird's and Corvette's but the 6th gear is .50:1 which is a very, very low ratio. The VH might be able to handle it but the Cobra T56 has around a .68:1 ratio, which would work out a little better.
On top of better gearing, they are very strong and there are many companies out there that can build them up even stronger. Lastly, they have a removable bell housing, which would make for a slightly easier setup.
