Does anybody know what that is circle in green on the block of a ka24de?

it is were one of the heater hoses connects to the block although my engine didnt have that fitting on it, it is the return or sending for the heater core

Since i don't have a heater or a/c system, could i just plug it off?

nope, you could bypass the heater core but you would still need a hose there

loop the hose to your other heater hose inlet on the intake side.
