if you have a dead cylinder, its not getting fuel, air or spark. that or it has no compression. Start with the spark test, make sure the plug is arcing and the wire is connected.
then look for a dead fuel injector. you can do this by "wiggling" the connector on top of the injector or removing/replacing it to look for an easy solution in a bad connection. If that doesnt work and the wiring looks good, pop the injector out and swap it with one from another cylinder (Its a simple procedure but requires effort and care so as not to damage the injectors when removing them). If the dead cylinder follows the injector from #3, assume that the injector has gone and needs to be replaced.
If #3 cyl is still dead with a known good injector, do a compression test. odds are against this being the culprit if only one cylinder is effected.
