To test the mechanical functionality, you can take off your interior door panel and unplug any and all wiring going to the door lock assembly. Now it's just a manual lock. Try and use the key to lock and unlock that. You will either be able to lock it up or you'll find some sort of physical interference such as the bar leading over the the interior manual lock switch. If you can lock it, then maybe the electrical lock mechanism is grounding out somewhere like in the hinge area of the door.
Oh, have you tried your interior manual lock switch? I mean, can you lock it by hand while you're inside the car? Maybe try that first, then what I said about unplugging and trying with the key. Then roll up the windows, lock it by hand, hold the handle up and shut the door. Just on one side at a time, you then need to verify you can unlock it so don't lock yourself out of both sides yet.
