Simple question, should the transmission fluid be checked with the engine running or off?

I'm getting a proper crosshatched area reading with the transmission warmed up and the engine OFF, but when I turn the engine ON to check the fluid it drops to the low area of the dipstick. Here's the kicker, I just removed about a quart of fluid because of this issue and now I've got this confusion :wtf2: . Bizarre.

So engine off or on with the car warmed up properly?
