Could I install a high-powered 6.5 speaker in that hole, and amp it mono?

I was thinking about getting a small amp and putting an actual sub there like the bose setup, but I already have a dual 10" bandpass box.

What are you hoping to gain with this 6.5" speaker? You said you have a 10" dual bandpass box, which should be giving you ample bass unless they aren't good subs or are underpowered. I'm guessing your desire for the 6.5" speaker is for stronger mid-bass response. The front bose aren't real capable at less than 100Hz. Thats' going to move you sound stage to the back of the car instead of the dashboard where you want it. If you consider yourself an audiophile you wouldn't want this. If you are a bass-head (and there's nothing wrong with that) then go for it. If you did put a speaker back there it should be an infinite baffle type or mounted in the proper enclosure.My suggestion for the best sound would be to look at upgrading your front components. Better 6.5"'s up front will make a big difference in the overall sound quality and staging.

yeah dont mess with the rear deck spot. in fact leavin that hole open will aloow air to pass into the cabin of the car giving you cleaner sounding bass from your subs.

Good point blanz. I just recommended last week to a guy that he should remove the Bose sub so the after sub bass can pass through the void.
