I am buying a 1991 300zx NA tomorrow, and I'm very new to all of this. I have a strong technical background, but almost no experience with cars.

With my DD, I can connect my laptop to an OBDII port and read/reset the codes via a serial terminal, but beyond that I have never used any sort of vehicle interface software. What software does everyone use? Bonus points if it runs on Linux.

I have seen a couple mentioned (conzult and nistune), but I'm not really sure which is used for what, or if there are better alternatives available. My goal is to be able to read as many sensors as possible, in order to diagnose problems, and eventually tune my ECU (so I'll also want something to read/write the EPROM).

Looking forward to hearing from you all!

Conzult is the fault reader primarily and Nistune that plus some tuning capabilites.
Realize that all Z32's except the 1996 version are OBD1 systems, and where '96 through '99 ('97 -'99 are Japan-only) goes OBD2.
The Z32 ECU's are not programmable OEM, a socketed ECU and aftermarket chip plus controller are necessary and further wrangling, all determined by your needs and any upgrades and modifications...basically Haltech is the best but costliest method for complete access and control of the engine, no other similarly capable options exist to my knowledge...yet.
